Within the STEM disciplines, it is common to have a committee that publishes a list of the major open research questions which the discipline should focus on within the next 5-10 years. A factor in discussions of what makes a discipline include having an area of research that is, by definition, within the scope of the discipline. Within those areas of research, there are major questions that the field works to answer; questions that change over course of years as research proceeds and new technology gets introduced.
